The strongest commercial choice is the one that performs consistently under real processing conditions.<\/p>\n<p><!-- lc-qa-end --><\/p>\n<p>\u805a\u5408\u9176\u94fe\u5f0f\u53cd\u5e94\uff08PCR\uff09\u662f\u4e00\u79cd\u5206\u5b50\u751f\u7269\u5b66\u6280\u672f\uff0c\u7528\u4e8e\u6269\u589e\u7279\u5b9a\u7684 DNA \u7247\u6bb5\uff0c\u88ab\u89c6\u4e3a\u751f\u7269\u4f53\u5916\u7684 DNA \u590d\u5236\uff0c\u5176\u4e2d\u5fae\u91cf\u7684 DNA \u7247\u6bb5\u88ab\u6269\u589e\u5230\u5f88\u5bb9\u6613\u8bc6\u522b\u548c\u8fa8\u8ba4\u7684\u7a0b\u5ea6\u3002<\/p>\n<p>\u5728 PCR \u53cd\u5e94\u4e2d\uff0c\"\u9176 \"\u8d77\u7740\u81f3\u5173\u91cd\u8981\u7684\u4f5c\u7528\uff0c\u6bcf\u79cd \"\u9176 \"\u90fd\u6709\u4e0d\u540c\u7684\u6d3b\u6027\u548c\u529f\u80fd\u3002\u90a3\u4e48\uff0c\u9176\u6709\u54ea\u4e9b\u79cd\u7c7b\uff1f\u5728 IVD \u9886\u57df\u53c8\u6709\u54ea\u4e9b\u5e94\u7528\uff1f<\/p>\n<p><strong>DNA \u805a\u5408\u9176<\/strong><\/p>\n<p>\u5b9a\u4e49\uff1a\u5b83\u662f\u4e00\u79cd\u4ee5 DNA \u4e3a\u6a21\u677f\uff0c\u4ece DNA \u7684 5 \u7aef\u590d\u5236\u5230 3 \u7aef\uff0c\u5e76\u50ac\u5316\u5e95\u7269 dNTP \u5206\u5b50\u805a\u5408\u5f62\u6210\u5b50 DNA \u7684\u9176\u3002<\/p>\n<p>\u4e3b\u8981\u6d3b\u52a8\u548c\u4f5c\u7528\uff1a\u50ac\u5316 DNA \u5408\u6210\u30021\u3001\u805a\u5408\u30022\u30013\u3001\u21925\u3001\u5916\u5207\u9176\u6d3b\u6027--\u6821\u5bf9\u4f5c\u7528 3\u30015\u3001\u21923\u3001\u5916\u5207\u9176\u6d3b\u6027--\u5207\u9664\u4fee\u590d\u4f5c\u7528 4\u3001\u7126\u78f7\u9178\u6c34\u89e3\u4f5c\u7528 5\u3001\u7126\u78f7\u9178\u4ea4\u6362\u3002<\/p>\n<p>IVD \u9886\u57df\u5e38\u89c1\u7684 DNA \u805a\u5408\u9176\u5305\u62ecTaq\u70ed\u542f\u52a8\u9176\uff0c\u5728\u53cd\u5e94\u4f53\u7cfb\u52a0\u70ed\u523070\u2103\u4e4b\u524d\uff0c\u5229\u7528\u5316\u5b66\u4fee\u9970\u5c01\u95edTaq\u9176\u7684\u6d3b\u6027\uff0c\u4ece\u800c\u6291\u5236\u975e\u7279\u5f02\u6027\u6269\u589e\u3002\u9ad8\u4fdd\u771f DNA \u805a\u5408\u9176\uff0c\u5305\u542b\u805a\u5408\u4e2d\u5fc3\u548c\u88c2\u89e3\u4e2d\u5fc3\uff0c\u805a\u5408\u4e2d\u5fc3\u5177\u6709 5\u2032-3\u2032 \u805a\u5408\u9176\u6d3b\u6027\uff0c\u8d1f\u8d23\u805a\u5408\uff1b\u88c2\u89e3\u4e2d\u5fc3\u5177\u6709 3\u2032-5\u2032 \u5916\u5207\u9176\u6d3b\u6027\uff08\u53c8\u79f0\u6821\u5bf9\u6d3b\u6027\uff09\uff0c\u8d1f\u8d23\u5c06\u672a\u914d\u5bf9\u7684\u6838\u82f7\u9178\u5207\u9664\u3002(\u5916\u5207\u9176\u6d3b\u6027\uff08\u53c8\u79f0\u6821\u5bf9\u6d3b\u6027\uff09\uff0c\u8d1f\u8d23\u5207\u9664\u672a\u914d\u5bf9\u7684\u6838\u82f7\u9178\u3002<\/p>\n<p><strong>\u9006\u8f6c\u5f55\u9176<\/strong><\/p>\n<p>\u5b9a\u4e49\uff1a\u4e00\u79cd\u9176\uff0c\u4ee5 RNA \u4e3a\u6a21\u677f\uff0c\u5728 5\u2032-3\u2032 \u65b9\u5411\u5408\u6210\u4e0e RNA \u6a21\u677f\u4e92\u8865\u7684 DNA \u5355\u94fe\u3002<\/p>\n<p>\u4e3b\u8981\u6d3b\u52a8\u548c\u4f5c\u7528\uff1a 1. RNA \u5f15\u5bfc\u7684 DNA \u805a\u5408\u9176\u6d3b\u6027\u30022.RNase H \u6d3b\u6027 3.DNA \u5f15\u5bfc\u7684 DNA \u805a\u5408\u9176\u6d3b\u6027\u3002<\/p>\n<p>\u5e94\u7528\u4e8e IVD \u9886\u57df\uff1a1\u3001\u53cd\u8f6c\u5f55\u805a\u5408\u9176\u94fe\u53cd\u5e94\uff08RT-PCR\uff09 2\u3001cDNA \u6587\u5e93\u6784\u5efa 3\u3001RNA \u6d4b\u5e8f 4\u3001RT-qPCR \uff0c2019 \u5168\u65b0\u7687\u51a0\u6838\u9178\u68c0\u6d4b\u914d\u7f6e\u7684\u7cfb\u7edf\u8bd5\u5242\u4e2d\uff0c\u5fc5\u7136\u4f1a\u52a0\u5165\u53cd\u8f6c\u5f55\u9176\u3002\u8fd9\u662f\u56e0\u4e3a RNA \u75c5\u6bd2\u5fc5\u987b\u9006\u8f6c\u5f55\u4e3a DNA \u624d\u80fd\u8fdb\u884c PCR \u53cd\u5e94\u3002<\/p>\n<p><strong>\u6838\u7cd6\u6838\u9178\u805a\u5408\u9176<\/strong><\/p>\n<p>\u5b9a\u4e49\uff1a\u4e00\u79cd\u4ee5 DNA \u6216 RNA \u94fe\u4e3a\u6a21\u677f\u7684\u805a\u5408\u9176\uff0c\u56e0\u4e3a\u5728\u7ec6\u80de\u5185\u5177\u6709 DNA \u8f6c\u5f55\u7684\u9057\u4f20\u4fe1\u606f\uff0c\u53c8\u79f0\u8f6c\u5f55\u9176\u3002<\/p>\n<p>\u4e3b\u8981\u6d3b\u52a8\u548c\u4f5c\u7528\uff1a1\u3001\u4ee5 DNA \u4e3a\u6a21\u677f\uff1b2\u3001\u901a\u8fc7\u805a\u5408\u53cd\u5e94\u50ac\u5316\u6838\u9178\u7684\u5408\u6210\uff1b3\u3001\u4f7f\u6838\u82f7\u9178\u5f62\u6210 3,\u21925,-\u78f7\u9178\u4e8c\u916f\u952e\uff1b4\u3001\u6839\u636e\u78b1\u57fa\u4e92\u8865\u914d\u5bf9\u539f\u7406\uff0c\u6cbf 5\u2032-3\u2032 \u65b9\u5411\u8f6c\u5f55\u6a21\u677f\u5e8f\u5217\u3002<\/p>\n<p>IVD \u9886\u57df\u5e38\u89c1\u7684 RNA \u805a\u5408\u9176\u5305\u62ec T7 RNA \u805a\u5408\u9176\uff0c\u5b83\u4e13\u95e8\u50ac\u5316 5'\u21923\u2032 \u65b9\u5411\u7684 RNA \u5f62\u6210\u8fc7\u7a0b\u3002\u76ee\u524d\u5728\u4f53\u5916\u8bca\u65ad\u9886\u57df\uff0cSAT \u6280\u672f\u5c31\u662f\u5e94\u7528 T7 RNA \u805a\u5408\u9176\u8fdb\u884c RNA \u6269\u589e\u3002\u8be5\u6280\u672f\u5df2\u5e7f\u6cdb\u5e94\u7528\u4e8e\u75c5\u539f\u4f53\u68c0\u6d4b\u3001\u8840\u6db2\u75c5\u6bd2\u7b5b\u67e5\u548c\u73af\u5883\u5fae\u751f\u7269\u68c0\u6d4b\u3002<\/p>\n<p><strong>\u86cb\u767d\u9176 K<\/strong><\/p>\n<p>\u5b9a\u4e49\u86cb\u767d\u9176 K \u662f\u4e00\u79cd\u4ece\u767d\u8272\u5ff5\u73e0\u83cc\u4e2d\u5206\u79bb\u51fa\u6765\u7684\u5f3a\u529b\u86cb\u767d\u6c34\u89e3\u9176\uff0c\u662f\u63d0\u53d6 DNA \u7684\u5173\u952e\u8bd5\u5242\u3002<\/p>\n<p>\u4e3b\u8981\u529f\u80fd\uff1a\u964d\u89e3\u819c\u86cb\u767d\u548c DNA \u7ed3\u5408\u86cb\u767d\uff0c\u4fc3\u8fdb DNA \u7684\u6e38\u79bb\u3002\u5728 IVD \u9886\u57df\u7684\u5e94\u7528\uff1a\u7528\u4e8e\u5206\u79bb\u548c\u63d0\u53d6\u8d28\u7c92\u3001\u57fa\u56e0\u7ec4 DNA\u3001RNA\u3002\u5728\u75c5\u6bd2\u6838\u9178\u68c0\u6d4b\u4e2d\uff0c\u86cb\u767d\u9176 K \u53ef\u88c2\u89e3\u548c\u706d\u6d3b\u75c5\u6bd2\u58f3\u86cb\u767d\uff0c\u540c\u65f6\u91ca\u653e\u75c5\u6bd2\u57fa\u56e0\u7ec4\uff0c\u5b8c\u6210\u6838\u9178\u63d0\u53d6\u3002<\/p>\n<p><strong>UDG \u9176<\/strong><\/p>\n<p>\u5b9a\u4e49\uff1a\u522b\u540d\u5c3f\u5627\u5576-DNA \u7cd6\u57fa\u5316\u9176\uff0c\u53c8\u79f0\u5c3f\u5627\u5576-N-\u7cd6\u57fa\u5316\u9176\u6216 UNG \u9176\u3002\u5b83\u662f\u5f62\u6210 DNA \u548c RNA \u5355\u4f53\u5e76\u7f16\u7801\u9057\u4f20\u4fe1\u606f\u7684\u5316\u5b66\u7ed3\u6784\u3002<\/p>\n<p>\u529f\u80fd\uff1a\u6709\u6548\u63a7\u5236 PCR \u6b8b\u7559\u6c61\u67d3\u3002\u5e94\u7528\u4e8e IVD\uff1a\u7528\u4e8e\u4e34\u5e8a\u68c0\u6d4b\u7684 PCR \u8bd5\u5242\u542b\u6709 UDG \u9176\uff0c\u53ef\u9632\u6b62\u6c61\u67d3\u3002<\/p>\n<p><b>\u5185\u5207\u9176<\/b><\/p>\n<p>\u5b9a\u4e49\uff1a\u4e00\u7c7b\u80fd\u8bc6\u522b\u7279\u5b9a\u6838\u82f7\u9178\u5e8f\u5217\u5e76\u5728\u6bcf\u6761\u6838\u82f7\u9178\u94fe\u7684\u7279\u5b9a\u4f4d\u70b9\u4e0a\u88c2\u89e3\u4e24\u4e2a\u8131\u6c27\u6838\u82f7\u9178\u4e4b\u95f4\u7684\u78f7\u9178\u4e8c\u916f\u952e\u7684\u9176\uff0c\u79f0\u4e3a\u9650\u5236\u9176\u3002<\/p>\n<p>\u4f5c\u7528\uff1a\u6709 3 \u79cd\u7c7b\u578b\uff1a\u7c7b\u578b 1\uff1a\u65e2\u6709\u4fee\u9970\u53c8\u6709\u8bc6\u522b\u88c2\u89e3\uff0c\u88c2\u89e3\u4f4d\u70b9\u79bb\u8bc6\u522b\u5e8f\u5217\u8f83\u8fdc\uff1b\u7c7b\u578b 2\uff1a\u53ea\u6709\u8bc6\u522b\u88c2\u89e3\uff0c\u88c2\u89e3\u4f4d\u70b9\u5c31\u662f\u8bc6\u522b\u5e8f\u5217\uff1b\u7c7b\u578b 3\uff1a\u65e2\u6709\u4fee\u9970\u53c8\u6709\u8bc6\u522b\u88c2\u89e3\uff0c\u88c2\u89e3\u4f4d\u70b9\u4e0e\u8bc6\u522b\u5e8f\u5217\u8f83\u8fd1\u3002<\/p>\n<p>\u5728 IVD \u9886\u57df\u7684\u5e94\u7528\uff1a1\uff0c\u57fa\u56e0\u5b9a\u4f4d\u548c\u57fa\u56e0\u5206\u79bb\uff1b2\uff0cDNA \u5206\u5b50\u78b1\u57fa\u5e8f\u5217\u5206\u6790\uff1b3\uff0c\u57fa\u56e0\u5de5\u7a0b\u7f16\u8f91\uff1b4\uff0cDNA \u7269\u7406\u56fe\u8c31\u7684\u5f62\u6210\u3002<\/p>\n<p><strong>\u8131\u94a9\u9176<\/strong><\/p>\n<p>\u5b9a\u4e49\uff1a\u5229\u7528 ATP \u6c34\u89e3\u63d0\u4f9b\u7684\u80fd\u91cf\uff0c\u89e3\u9664\u53cc\u94fe DNA \u6838\u82f7\u9178\u914d\u5bf9\u5f62\u6210\u7684\u6c22\u952e\uff0c\u5f62\u6210\u5355\u94fe DNA\u3002<\/p>\n<p>\u4f5c\u7528\uff1a\u5728 DNA \u6216 RNA \u590d\u5236\u8fc7\u7a0b\u4e2d\u50ac\u5316 DNA \u6216 RNA \u7684\u89e3\u65cb\u3002<\/p>\n<p>\u5e94\u7528\u4e8e IVD \u9886\u57df\uff1a\u7b49\u6e29\u6269\u589e\u6280\u672f\uff0c\u5229\u7528\u70ed\u7a33\u5b9a\u89e3\u805a\u9176\u4ee3\u66ff qPCR \u4e2d\u7684\u70ed\u53d8\u6027\u8fc7\u7a0b\uff0c\u89e3\u5f00\u53cc\u94fe\uff0c\u5ef6\u957f\u6269\u589e\u65f6\u95f4\u3002\u8fd9\u79cd\u6280\u672f\u4e0d\u6613\u4ea7\u751f\u975e\u7279\u5f02\u6027\u6269\u589e\uff0c\u4e5f\u907f\u514d\u4e86 PCR \u5faa\u73af\u4e2d\u5347\u6e29\u548c\u964d\u6e29\u7684\u8fc7\u7a0b\uff0c\u53ea\u9700\u8981\u7b80\u5355\u7684\u68c0\u6d4b\u8bbe\u5907\u3002<\/p>\n<p><strong>\u4fee\u9970\u9176<\/strong><\/p>\n<p>\u5b9a\u4e49\u4e00\u79cd\u50ac\u5316\u7a00\u6709\u78b1\u57fa\u63ba\u5165 RNA \u6216 DNA \u7684\u9176\uff0c\u6216\u4e00\u79cd\u4fee\u9970\u539f\u59cb\u78b1\u57fa\u4ee5\u9632\u6b62\u88ab\u9650\u5236\u6027\u5185\u5207\u9176\u7834\u574f\u7684\u9176\u3002<\/p>\n<p>\u4f5c\u7528\uff1a\u5bf9\u5176\u4ed6\u9176\u7684\u7ed3\u6784\u8fdb\u884c\u5171\u4ef7\u4fee\u9970\uff0c\u4f7f\u5176\u5728\u9ad8\u6d3b\u6027\u5f62\u5f0f\u548c\u76f8\u5bf9\u4f4e\u6d3b\u6027\u5f62\u5f0f\u4e4b\u95f4\u76f8\u4e92\u8f6c\u5316\u3002<\/p>\n<p>\u5e94\u7528\u4e8e IVD\uff1a5mC\/m6A \u6838\u9178\u4fee\u9970\u9176\u6d3b\u6027\u68c0\u6d4b HTMR \u65b9\u6cd5\u4e3a\u53d1\u73b0\u548c\u8bc4\u4f30\u9488\u5bf9 DNA\/RNA \u7532\u57fa\u5316\u7684\u5c0f\u5206\u5b50\u4fee\u9970\u5242\u63d0\u4f9b\u4e86\u6709\u529b\u8bc1\u636e\u3002<\/p>\n<p><strong>\u7126\u78f7\u9178\u9176<\/strong><\/p>\n<p>\u5b9a\u4e49\uff1a\u50ac\u5316\u4e00\u5206\u5b50\u7126\u78f7\u9178\u8f6c\u5316\u4e3a\u4e24\u5206\u5b50\u78f7\u9178\u79bb\u5b50\u7684\u9176\u3002<\/p>\n<p>\u529f\u80fd\uff1a\u70ed\u7a33\u5b9a\u6027\u65e0\u673a\u7126\u78f7\u9178\u9176\u5728 100\u00b0C \u4e0b\u52a0\u70ed 4 \u5c0f\u65f6\u540e\u4ecd\u5177\u6709 100% \u6d3b\u6027\uff0c\u56e0\u6b64\u53ef\u7528\u4e8e PCR \u53cd\u5e94\uff0c\u89e3\u9664\u751f\u6210\u7684\u65e0\u673a\u7126\u78f7\u9178\u9176\u5bf9 PCR \u7684\u6291\u5236\uff0c\u4fc3\u8fdb DNA \u590d\u5236\u3002<\/p>\n<p><b>taq \u6297\u4f53<\/b><\/p>\n<p>\u5b9a\u4e49\u6297 Taq DNA \u805a\u5408\u9176\u7684\u6297\u4f53\u3002<\/p>\n<p>\u529f\u80fd\uff1a\u5728 PCR \u6269\u589e\u8fc7\u7a0b\u4e2d\uff0cTaq \u6297\u4f53\u4e0e Taq \u9176\u7ed3\u5408\uff0c\u5728\u9ad8\u6e29\u53d8\u6027\u524d\u6291\u5236 DNA \u805a\u5408\u9176\u7684\u6d3b\u6027\uff0c\u5e76\u80fd\u6709\u6548\u6291\u5236\u5f15\u7269\u7684\u975e\u7279\u5f02\u6027\u9000\u706b\u548c\u5f15\u7269\u4e8c\u805a\u4f53\u5728\u4f4e\u6e29\u4e0b\u5f15\u8d77\u7684\u975e\u7279\u5f02\u6027\u6269\u589e\u3002<\/p>\n<p>\u5728 IVD \u4e2d\u7684\u5e94\u7528\uff1aTaq \u6297\u4f53\u7684\u9ad8\u5c01\u95ed\u6027\u53ef\u63d0\u9ad8\u65b0\u578b\u51a0\u72b6\u75c5\u6bd2\u6838\u9178\u68c0\u6d4b\u8bd5\u5242\u76d2\u7684\u68c0\u6d4b\u7075\u654f\u5ea6\uff0c\u6709\u52a9\u4e8e\u65b0\u578b\u51a0\u72b6\u75c5\u6bd2\u7684\u68c0\u6d4b\u3002<\/p>\n<p>\u6807\u51c6\u7684 PCR \u8fc7\u7a0b\u5206\u4e3a\u4e09\u4e2a\u6b65\u9aa4\uff1aDNA \u53d8\u6027\u3001\u9000\u706b\u548c\u5ef6\u4f38\u3002\u706d\u6d3b\u53cd\u5e94\u53ef\u5728 PCR \u5f00\u59cb\u65f6\u8fdb\u884c\uff0c\u5347\u6e29\u81f3 50\u00b0C 10 \u5206\u949f\u6216 95\u00b0C 2 \u5206\u949f\uff0cUDG \u9176\u53ef\u6d88\u9664\u53cd\u5e94\u4f53\u7cfb\u4e2d\u6c61\u67d3 DNA \u4ea7\u751f\u7684\u6269\u589e\u3002\u968f\u540e\uff0c\u6a21\u677f DNA \u7ecf\u9ad8\u6e29\u53d8\u6027\u4e3a\u5355\u94fe DNA\u3002\u5728 DNA \u805a\u5408\u9176\u548c\u9002\u5f53\u6e29\u5ea6\u4e0b\uff0c\u4e24\u6761\u5f15\u7269\u5206\u522b\u4e0e\u4e24\u6761\u6a21\u677f DNA \u94fe\u9000\u706b\u3002Taq \u6297\u4f53\u4e0e Taq \u9176\u7ed3\u5408\uff0c\u6291\u5236 DNA \u805a\u5408\u9176\u7684\u6d3b\u6027\uff0c\u4ece\u800c\u6291\u5236\u4f4e\u6e29\u4e0b\u7684\u975e\u7279\u5f02\u6027\u6269\u589e\u3002\u5728 PCR
