A colesterol oxidase (EC1.1.3.6 COD) est\u00e1 sendo cada vez mais reconhecida por sua fun\u00e7\u00e3o no processamento de alimentos, em testes m\u00e9dicos, no controle biol\u00f3gico de pragas e em outros campos, e tem demonstrado grande potencial de aplica\u00e7\u00e3o.
\n\u00c9 uma enzima essencial no processo de metabolismo do colesterol, dependente da coenzima FAD, e pode catalisar especificamente a convers\u00e3o do colesterol em colest-4-eno-3-ona (4-colesterol-3-ona). A E. coli recombinante que produz COD foi usada como objeto de pesquisa, com o objetivo de desenvolver e aplicar COD. Foram realizadas pesquisas sobre a separa\u00e7\u00e3o e a extra\u00e7\u00e3o da CQO, suas propriedades enzim\u00e1ticas e a aplica\u00e7\u00e3o da enzima. A CQO foi isolada por meio de um processo de purifica\u00e7\u00e3o que incluiu tratamento ultrass\u00f4nico de E. coli recombinante, salga de 70% (NH_4)_2SO_4, cromatografia de troca i\u00f4nica CM Sepharose F.F. e filtra\u00e7\u00e3o em gel fino Sephadex G-75. A DQO extra\u00edda e purificada tinha uma atividade enzim\u00e1tica de 47,3 U\/mg, um m\u00faltiplo de purifica\u00e7\u00e3o de 45 vezes e uma taxa de recupera\u00e7\u00e3o da atividade enzim\u00e1tica de 21%.<\/p>\n
