La cholest\u00e9rol oxydase (EC1.1.3.6 COD) est de plus en plus reconnue pour son r\u00f4le dans la transformation des aliments, les tests m\u00e9dicaux, la lutte biologique contre les parasites et d'autres domaines, et elle a montr\u00e9 un grand potentiel d'application.
\nIl s'agit d'une enzyme cl\u00e9 dans le processus du m\u00e9tabolisme du cholest\u00e9rol, qui d\u00e9pend de la coenzyme FAD, et qui peut catalyser sp\u00e9cifiquement la conversion du cholest\u00e9rol en cholest\u00e9rol-4-\u00e8ne-3-one (4-cholest\u00e9rol-3-one). L'E. coli recombinant produisant la DCO a \u00e9t\u00e9 utilis\u00e9 comme objet de recherche, dans le but de d\u00e9velopper et d'appliquer la DCO. Les recherches ont port\u00e9 sur la s\u00e9paration et l'extraction de la DCO, ses propri\u00e9t\u00e9s enzymatiques et l'application de l'enzyme. La DCO a \u00e9t\u00e9 isol\u00e9e \u00e0 l'aide d'un processus de purification comprenant un traitement ultrasonique de l'E. coli recombinant, un salage 70% (NH_4)_2SO_4, une chromatographie d'\u00e9change d'ions CM Sepharose F.F. et une filtration sur gel Sephadex G-75 Fine. La DCO extraite et purifi\u00e9e avait une activit\u00e9 enzymatique de 47,3 U\/mg, un multiple de purification de 45 fois et un taux de r\u00e9cup\u00e9ration de l'activit\u00e9 enzymatique de 21%.<\/p>\n
