Li Ping a utilis\u00e9 la m\u00e9thode du spectrophotom\u00e8tre p-nitroph\u00e9nol, dans l'exp\u00e9rience d'ajout de la \u03b2-glucosidase d'Aspergillus niger dans la solution de p-nitroph\u00e9nol -\u03b2-glucoside ( pNPG ) pour l'hydrolyse enzymatique, et la valeur de l'absorbance a \u00e9t\u00e9 mesur\u00e9e \u00e0 une longueur d'onde ultraviolette de 400 nm. Selon la valeur d'absorbance mesur\u00e9e, la quantit\u00e9 restante du substrat naringine peut \u00eatre calcul\u00e9e, puis l'activit\u00e9 naringinase peut \u00eatre obtenue par calcul. Wang Weina et al. ont utilis\u00e9 une m\u00e9thode de Davis am\u00e9lior\u00e9e, ont pris la naringine comme substrat et ont ajout\u00e9 de la naringinase produite par fermentation liquide d'Aspergillus niger FFCC 848 pour l'hydrolyse enzymatique. L'absorbance a \u00e9t\u00e9 mesur\u00e9e sous une lumi\u00e8re ultraviolette de 420 nm, puis l'activit\u00e9 enzymatique a \u00e9t\u00e9 calcul\u00e9e. La m\u00e9thode du spectrophotom\u00e8tre de Davis pour mesurer l'activit\u00e9 enzymatique est simple \u00e0 utiliser et rapide. Cette m\u00e9thode est donc largement utilis\u00e9e pour la d\u00e9tection en temps r\u00e9el de la production de naringinase par fermentation et de l'activit\u00e9 de la naringinase immobilis\u00e9e.<\/h4>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4580\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-42.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h3 class=\"wp-block-heading\">1-2 Chromatographie liquide \u00e0 haute performance<\/h3>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4579\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-41.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h4 class=\"wp-block-heading\">Le principe de la chromatographie liquide \u00e0 haute performance (CLHP) est que la CLHP peut d\u00e9terminer avec pr\u00e9cision et analyser quantitativement le substrat naringine, le produit interm\u00e9diaire purunine et le produit final naring\u00e9nine pendant tout le processus d'hydrolyse, puis calculer l'activit\u00e9 enzymatique. Chen Yuelong et d'autres ont utilis\u00e9 la CLHP pour d\u00e9terminer le temps de r\u00e9tention de la naringine, de l'arbutine, de la myric\u00e9tine et de leurs produits, et ont r\u00e9alis\u00e9 une s\u00e9paration compl\u00e8te avec une r\u00e9solution sup\u00e9rieure \u00e0 1,5, et ont contr\u00f4l\u00e9 efficacement l'effet de la naringinase sur de multiples glycosides dans le processus d'hydrolyse, tout en d\u00e9terminant avec pr\u00e9cision l'activit\u00e9 enzymatique. La m\u00e9thode CLHP pr\u00e9sente les avantages d'une grande pr\u00e9cision et peut \u00e9viter l'interf\u00e9rence des impuret\u00e9s, mais compar\u00e9e \u00e0 la m\u00e9thode Davis, cette m\u00e9thode a un temps de d\u00e9tection plus long et une op\u00e9ration plus compliqu\u00e9e.<\/h4>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4578\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-40.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h3 class=\"wp-block-heading\">1-3 M\u00e9thode am\u00e9lior\u00e9e de d\u00e9termination de l'activit\u00e9 enzymatique de la naringinase<\/h3>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4577\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-39.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h4 class=\"wp-block-heading\">En utilisant la m\u00e9thode Davis am\u00e9lior\u00e9e : 0,8 ml de 0,8 g-L<sup>-1<\/sup>\u00a0Une solution de naringine pr\u00e9par\u00e9e avec un tampon pH 4,5 a \u00e9t\u00e9 ajout\u00e9e au syst\u00e8me r\u00e9actionnel contenant 40 mg de naringinase immobilis\u00e9e (ou 0,2 mL de naringinase libre). Le placer dans un bain-marie \u00e0 temp\u00e9rature constante de 50 \u2103 pendant 30 minutes. Pr\u00e9lever 0,1 mL de la solution d'hydrolyse enzymatique r\u00e9actionnelle et ajouter 5 mL de di\u00e9thyl\u00e8ne glycol (fraction volumique de 90%) et 0,1 mL de solution de NaOH (4 mol-L<sup>-1<\/sup>) dans des tubes et laisser reposer pendant 10 minutes apr\u00e8s les avoir m\u00e9lang\u00e9s uniform\u00e9ment. Mesurer l'absorbance de la solution m\u00e9lang\u00e9e \u00e0 420 nm.<\/h4>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4576\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-38.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h4 class=\"wp-block-heading\">La d\u00e9finition de l'activit\u00e9 de l'enzyme naringinase (U) : \u00e0 une condition de pH 4,5, 50 \u2103, 1 unit\u00e9 d'activit\u00e9 enzymatique est la quantit\u00e9 d'enzyme n\u00e9cessaire pour d\u00e9grader 1 \u03bcg de naringine par minute. Le rapport d'activit\u00e9 enzymatique ( U-g<sup>-1<\/sup>\u00a0ou U-mL<sup>-1<\/sup>) est d\u00e9finie comme suit : \u00e0 pH 4,5, 50 \u2103, activit\u00e9 enzymatique pr\u00e9sent\u00e9e par 1 g de naringinase immobilis\u00e9e (ou 1 mL de naringinase libre). Pr\u00e9paration de la solution enzymatique de naringinase<\/strong><\/h2>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4572\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-34.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h4 class=\"wp-block-heading\">L'Aspergillus niger FFCC uv-11 conserv\u00e9 \u00e0 4 \u2103 a \u00e9t\u00e9 transf\u00e9r\u00e9 sur le milieu de culture, et les spores matures ont \u00e9t\u00e9 obtenues par culture \u00e0 temp\u00e9rature constante \u00e0 30 \u2103 pendant 96 h . Ensuite, les spores ont \u00e9t\u00e9 lav\u00e9es avec une solution saline normale st\u00e9rile avec une fraction de masse de 0,9%. La valeur de l'absorbance de la suspension de spores est mesur\u00e9e \u00e0 600 nm (valeur OD600), avec une solution saline st\u00e9rile sa valeur OD600 a \u00e9t\u00e9 ajust\u00e9e \u00e0 0,200. Ensuite, la suspension de spores avec la fraction volumique de 10% a \u00e9t\u00e9 inocul\u00e9e dans 100 ml de milieu de fermentation (fiole conique de 250 ml). Puis \u00e0 travers le filtre d'aspiration pour obtenir le liquide surnageant n\u00e9cessaire naringine enzyme fluide. Le r\u00e9frig\u00e9rer \u00e0 4 \u2103 pour une utilisation ult\u00e9rieure.<\/h4>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4571\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-33.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h2 class=\"wp-block-heading\"><strong>3. Pr\u00e9paration de l'enzyme Naringinase immobilis\u00e9e dans le chitosane<\/strong><\/h2>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4570\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-32.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h4 class=\"wp-block-heading\">Prendre 3,0 g de chitosane, avec 100 ml de fraction volumique de la solution d'acide ac\u00e9tique 2% a \u00e9t\u00e9 dissous pour pr\u00e9parer 0,3 g - L<sup>-1<\/sup>\u00a0La solution collo\u00efdale de chitosane a \u00e9t\u00e9 soniqu\u00e9e pendant 20 minutes afin d'\u00e9liminer les bulles d'air et de la laisser se dissoudre compl\u00e8tement. Les \u00e9tapes exp\u00e9rimentales sont les suivantes 0,1 g de Fe<sub>3<\/sub>O<sub>4<\/sub>\u00a0ont \u00e9t\u00e9 uniform\u00e9ment dispers\u00e9es dans une solution mixte contenant 40 ml d'\u00e9thanol anhydre, 10 ml d'eau distill\u00e9e et 1,2 ml d'eau ammoniaqu\u00e9e concentr\u00e9e. Sous une agitation de 300 r-min<sup>-1<\/sup>Des gouttelettes de solution contenant du Fe<sub>3<\/sub>O<sub>4<\/sub>\u00a0ont \u00e9t\u00e9 ajout\u00e9es au TEOS (0,4 ml) dispers\u00e9 dans 30 ml d'\u00e9thanol anhydre, et la r\u00e9action des nanoparticules \u00e0 base de silicium s'est d\u00e9roul\u00e9e pendant 12 heures \u00e0 temp\u00e9rature ambiante. Les produits ont \u00e9t\u00e9 lav\u00e9s plusieurs fois avec de l'\u00e9thanol anhydre et de l'eau distill\u00e9e pour obtenir des nanoparticules magn\u00e9tiques \u00e0 base de silice (MS), qui ont \u00e9t\u00e9 s\u00e9ch\u00e9es en vue d'une utilisation ult\u00e9rieure.<\/h4>\r\n\r\n\r\n\r\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" class=\"wp-image-4564\" src=\"https:\/\/longchangchemical.com\/wp-content\/uploads\/2021\/04\/image-26.png\" alt=\"\" \/><\/figure>\r\n\r\n\r\n\r\n<h4 class=\"wp-block-heading\">Les microsph\u00e8res de MSC ont \u00e9t\u00e9 pr\u00e9par\u00e9es par la m\u00e9thode de r\u00e9ticulation en \u00e9mulsion, comme le montre la figure 2. Les \u00e9tapes exp\u00e9rimentales \u00e9taient les suivantes : 0,125 g de nanoparticules de silice magn\u00e9tiques (MS) uniform\u00e9ment dispers\u00e9es dans 10 mL de solution de chitosane avec une fraction massique de 2,5% pr\u00e9par\u00e9e par le chitosane dissous dans une solution d'acide ac\u00e9tique avec une fraction volumique de 5,0%, puis ajouter la solution de chitosane dans une \u00e9mulsion contenant 60 mL de cire de paraffine liquide et 1,8 mL de twain 80 goutte \u00e0 goutte. Apr\u00e8s 30 minutes d'agitation au bain-marie \u00e0 40 \u2103 et \u00e0 800 r-min, la solution de chitosane est m\u00e9lang\u00e9e \u00e0 la cire de paraffine liquide.<sup>-1<\/sup>\uff0cadditionner 2,0 mL de glutarald\u00e9hyde et faire r\u00e9agir 1 h. Ensuite, le pH du syst\u00e8me a \u00e9t\u00e9 ajust\u00e9 \u00e0 9,0 avec 1 mol-L<sup>-1<\/sup>\u00a0NaOH, et la r\u00e9action a \u00e9t\u00e9 poursuivie pendant 1 h. Apr\u00e8s agitation, si la ph\u00e9nolphtal\u00e9ine ne vire pas au rouge, cela signifie qu'il n'y a pas d'OH- libre dans le MSCE ; prendre 3,0 mL du surnageant de lavage et ajouter 3,0 mL de 1,3 mol-L<sup>-1<\/sup>\u00a0thiosulfate de sodium et 1 ~2 gouttes de ph\u00e9nolphtal\u00e9ine, si la solution ne devient pas rouge apr\u00e8s agitation, cela signifie qu'il n'y a pas de groupes \u00e9poxy libres dans le MSCE. Apr\u00e8s la r\u00e9action, les microsph\u00e8res ont \u00e9t\u00e9 lav\u00e9es plusieurs fois avec une solution tampon jusqu'\u00e0 ce qu'il n'y ait plus de naringinase libre \u00e0 la surface des microsph\u00e8res.
